The luminous markers should be evenly applied with no bleed outside their outlines.

Run the movement through its motions. The seconds hand on a genuine automatic Rolex sweeps smoothly rather than ticking, and the rotor should wind smoothly with no grinding when you shake the watch gently. If you can, have a specialist open the caseback (or inspect through a case-opening tool) to confirm the calibre matches what's expected for the reference and production year.
